New York to Johannesburg (JFK to JNB) with South African Airways (although some SAA flights involve a stop in Dakar)
Atlanta to Johannesburg (ATL to JNB) with Delta Air Lines.
Washington Dulles to Johannesburg (IAD to JNB) with South African Airways.
All direct flights from the US to South Africa are to Johannesburg OR Tambo International Airport (JNB). If booking on onward flight from Johannesburg, note whether it's from Lanseria International Airport (HLA) or OR Tambo International Airport (JNB).

Note that the embassy is closed on South African as well as United States public holidays.
United States citizens require a visa to enter South Africa (if you're going on holiday you'll usually apply for a visitors visa). Make sure that you have at least one blank page in your passport, enough to pay for your needs during your visit to SA and a return flight ticket. There are South African missions in Chicago, Los Angeles and New York - the embassy is in Washington DC.
