In South Africa the railway transport systems are relatively underdeveloped, and so people tend to travel by plane (if you've got enough money) or by bus. If you are travelling a relatively long distance, then you'd be looking for an intercity bus (or as they call them in the USA, an interstate bus). Options include the Baz Bus, Citiliner, City to City, Greyhound, Intercape, APM and Translux buses. Bookings can be made via the bus hire company's websites, or via Computicket.
For shorter distance travelling one is looking at hiring a car, a taxi cab, minibus taxi or traveling on a local bus. Local buses and minibus taxis are usually the cheapest options. There are different booking systems in each town:

Cape Town Bus Bookings
The City of Cape Town has been implementing the MyCiTi bus service in phases. The first phase was set up for the 2010 Fifa World Cup, and transited passengers between Cape Town International Airport and the Civic Centre (in the city). In May 2011 a route between Table View and the Civic Centre was launched. This was later extended to service Dunoon, Joe Slovo Park, Melkbos, Atlantis and Montague Gardens. Then in 2013 a route between the Civic Centre and Salt River Railway Station was launched, which services Hout Bay, Camps Bay, Sea Point, Tamboerskloof, Orangezicht, District Six and Walmer Estate. Durban Bus Bookings

People Mover covers two main bus routes, the City Loop and the Beach Line.
The City Loop starts and ends at the Dr Pixley KaSeme Street transfer interchange, stopping near Durban City Hall, The Playhouse Theatre, Tourism Information Centre, Inkosi Albert Luthuli International Convention Centre, the Victoria street market and Durban's mosque and Cathedral.
The Beach Line runs along the Northern and Southern city beaches, starting at the Suncoast Casino and Entertainment World and ending at uShaka Marine World.
